3. GS Sequencer : 3.3 Starting a Sequencing Run : 3.3.3 Service Procedures
The first Instrument Procedure Wizard window (‘Choose a procedure’; see Figure 9) includes a choice called “Service”, where the following services can be selected:
3.3.3.1
Pre-wash
The Pre-wash service starts a pre-wash of the instrument, which must be carried out prior to each sequencing Run, using reagents and supplies provided in the Sequencing Kits as described in the Sequencing Method Manual. Selecting this option from either here or from the first Instrument Procedure Wizard window opens the ‘Start Pre-wash’ window of the Instrument Procedure Wizard (Figure 21).
3.3.3.2
Maintenance Wash
The Maintenance Wash service presents an instruction window (Figure 22) similar to the one above, and the Start button is enabled when all instrument sensors report that the instrument is ready. Clicking ‘Start’ launches a Maintenance Wash. The Maintenance Wash procedure is described in the GS FLX+ Instrument Owner’s Manual, and requires a GS FLX Maintenance Wash Kit, available separately.
3.3.3.3
Hardware Maintenance (Filter and Pump Tubing Replacement)
The Hardware Maintenance service disengages the two primary pumps to allow the operator to replace the inline filters and pump tubing (Figure 23). This maintenance step is required after every 25 Sequencing Runs, or 3 months, whichever comes first. The filter and pump tubing replacement procedure are described in the GS FLX+ Instrument Owner’s Manual. When you are finished with the procedure, check the box and click the Finish button.
3.3.3.4
Custom Script (Select an Instrument Run Script)
The Custom Script service opens the ‘Select an instrument Run script’ window (Figure 25) where a special .icl file can be specified for the Run. Some of these script files come preinstalled on the instrument and are used by Roche Representatives during service of your GS FLX+ Instrument.
